Itachi killed off the Uchiha Clan. Sasuke is bent on revenge. Exactly, Itachi was conspired by one of founders of Konohagakure and the founder of Akatsuki (the organization that Itachi is a part of..), Madara Uchiha. So you can't fully blame Itachi for the wiping most of the clan (leaving only Sasuke). Itachi killed off the rest of Uchiha Clan to get the sharigan (not so sure though).

He may have missed as well. Only cutting certain areas that weren't vital to live. They didn't explain that though. Soon you will see that it won't matter anyways, as Tsunade basically completely recovers using a special power. Also, about Itachi, From what I could tell, Itachi's motive to kill the clan was not fueled by gaining the Mangekyou Sharingan. Which is simply an advanced form of normal Sharingan, giving you the ability to move faster, and use unbeatable Genjutsu among other things, Quote: The Mangekyo Sharingan appears to react in different ways for different ninja. Itachi's Mangekyo Sharingan enabled him to use Tsukuyomi, Amaterasu, and Susano'o. Madara Uchiha's Mangekyo Sharingan appears to allow him to utilize a powerful space / time teleportation jutsu similar in principal to the Flying Thunder God Technique but without any requirements for use. And as they said, each one looks different. I'm not going to explain who has what, but you can figure it out for yourself as it might spoil alot. And please, if you know who they are don't blurt it out and don't tell why either, that's way too many spoilers for one page.
